Classification

Protein glycosylation can be divided into four categories according to the connection mode of amino acids and sugars: O-glycosylation, N-glycosylation, C-mannose glycosylation and GPI (glycosphatidyinositol) anchored connection.

 

1. N-linked glycosylation is through the reduction end of the sugar chain, N-acetylaminoglucose (GLC NAC) is connected to the N atom on some ASN side chain amide groups in the peptide chain. The ASN that can be connected to the sugar chain must be in the motif composed of asn-x-ser / Thr (x! = P) residues. The sugar is N-acetylglucosamine.


2. The structure of O-glycosylation is simpler than that of N-glycosylation. Generally, the sugar chain is relatively short, but there are more kinds than that of N-glycosylation. Ser and Thr can be glycosylated in the peptide chain, in addition to Tyrosine, Hydroxylysine and Hydroxyproline. The linked sites are the hydroxyl oxygen atoms on the side chains of these residues. The linked sugar is galactose or N-acetylgalactosamine (Gal & GalNAc) {or glucose / glucosamine (GLC / GlcNAc), mannose / mannosamine (Man / ManNAc), etc
